丹酚酸B对大鼠非酒精性脂肪性肝炎的影响(1)

【摘要】 目的 探讨丹酚酸B对大鼠非酒精性脂肪性肝炎的疗效。方法 雄性SD大鼠36只随机分组,对照组、模型组及丹酚酸B治疗组各12只,对照组给予普通饲料,其余各组均给予高脂饲料。于12周处死模型组。治疗组每天以浓度为1 mg/ml的丹酚酸B溶液20 ml/kg灌胃,同时继续高脂饲料喂养,对照组以20 ml/kg蒸馏水灌胃,同时继续普通饲料喂养。于实验第24周处死剩余实验动物。测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、三酰甘油(TG)、总胆固醇(TC)及肝组织丙二醛(MDA)含量,同时观察肝组织病理学改变,免疫组化法检测肝组织TIMP1的表达,RTPCR法检测肝组织TGFβ1mRNA的表达。结果 与正常对照组相比,模型组肝指数、血清ALT、AST、TG、TC及肝组织MDA水平显著升高(P<001),肝组织炎症活动度计分、TIMP1及TGFβ1mRNA的表达均显著升高(P<001)。丹酚酸B治疗组血清ALT、AST、TG、TC及肝组织MDA水平较模型组显著降低(P<001),肝组织炎症活动度计分、TIMP1及TGFβ1mRNA的表达均较模型组显著降低(P<001 或P<005)。结论 丹酚酸B对高脂饮食大鼠非酒精性脂肪性肝炎有一定的治疗作用。

【Abstract】 Objective To study the therapeutic role of Salvianolic acid B(SAB)in treating nonalcoholic stentohepatitis(NASH)in rats Methods Thirtysix male SD rats were randomly divided into 3 groups The normal control group (n=12)wse fed with normal diet.NASH model group (n=12)and therapeutic group(n=12)were fed with fat rich diet, The rats of NASH model group were killed at the 12 th week Therapeutic group was treated with SAB at the dose of 10 mg/kg from the 13 th week and finished at the 24 th week The serum ALT, AST, TG, TC and liver tissue MOD levels were measured, The degrees of hepatic histologic change were observed with HE, Tissue inhibitor of metalloproteinase one(TIMP1) was examined with immunohistochemistry, And the gene mRNA level of transforming growth factor beta one(TGFβ1) was deteced with reverse transcription polymerase chain reaction(RTPCR)Results Compared with the normal control group, the liver index, the serum AIT, AST, TG, TC and liver tissue MOD levels were increased significantly(P<001)in NASH model control group.The histological activity index(HAI) score, expressions of TIMP1and TGFβ1mRNA were increased significantly(P<001) Compared with NASH modeI control group, The above indexes in the therapeutic group were decreased significantly(P<005 or P<001)Conclusion SAB has some therapeutic role in treating NASH of rats ......
