黄芪多糖对脾虚湿困大鼠内分泌、免疫相关因子的影响(1)

     摘要 目的:研究黄芪多糖对脾虚湿困大鼠内分泌、免疫相关因子的影响,探讨黄芪多糖健脾祛湿机制。方法:高脂低蛋白饮食加力竭游泳法建立脾虚湿困大鼠模型。黄芪多糖干预后观察脾虚湿困大鼠体质量变化,检测血清总蛋白(TP)、球蛋白(GLB)含量变化,ELISA法检测血清垂体腺苷酸环化酶活化肽(PACAP)含量,免疫组化法检测十二指肠组织IL-6、IL-10表达。结果:与模型组比较,黄芪多糖高剂量组大鼠体质量增加(P<0.05);黄芪多糖中、高剂量组血清TP含量升高(P<0.05);黄芪多糖低、中、高剂量组GLB水平出现升高(P<0.01),血清PACAP水平降低(P<0.01),十二指肠组织IL-6及IL-10阳性面积百分比降低(P<0.01,P<0.05)。结论:黄芪多糖可调节脾虚湿困大鼠内分泌、免疫系统相关因子进而发挥健脾祛湿功效。

    Abstract Objective:To study effects of astragalus polysaccharides(APS)on endocrine and immune-related factors in rats with dampness stagnancy due to spleen deficiency, and to explore the mechanism of APS in invigorating spleen for eliminating dampness.Methods:Rat model with dampness stagnancy due to spleen deficiency was established by the high fat and low protein diet and exhaustive swimming method.After APS intervention, the change in body weight was observed, changes in serum levels of total protein(TP)and globulin(GLB)were determined, the content of pituitary adenylate cyclase activating peptide(PACAP)was determined by the ELISA method, and expressions of IL-6 and IL-10 in the duodenum were measured by an immunohistochemical method.Results:Compared with model group, the rats′ body weights in high dose APS group were increased(P<0.05); the serum TP levels in medium and high dose APS groups were increased(P<0.05); the GLB levels in the low, medium and high dose APS groups were increased(P<0.01), the serum PACAP levels were decreased(P<0.01), and positive area percentages of IL-6 and IL-10 in the duodenum were decreased(P<0.01, P<0.05).Conclusion:APS can regulate endocrine and immune-related factors in rats with dampness stagnancy due to spleen deficiency to exert the effects of invigorating spleen for eliminating dampness.

    黄芪是豆科多年生草本植物蒙古黄芪或膜荚黄芪干燥根,性温味甘,归脾、肺经,具有补气升阳、益卫固表、利水消肿等功效[1-2]。黄芪多糖是黄芪主要活性成分之一,可以调节免疫、抗炎、抗氧化,在防治糖尿病、肿瘤、神经性疾病等方面具有一定作用[3-4]。我们前期研究发现黄芪及其拆分组分可改善脾虚湿困大鼠一般状况,黄芪多糖效果最好[5]。本实验将从内分泌、免疫调节角度,探讨黄芪多糖改善脾虚湿困的机制,为脾虚湿困的机制研究及黄芪多糖临床应用提供依据。

    1 材料与方法

    1.1 材料

    1.1.1 动物 SPF级Wistar大鼠,48只,雌雄各半,4周龄,体质量100~120 g,购于北京维通利华实验动物技术有限公司,动物许可证编号:SCXK(京)2014-0004。大鼠在IVC独立送回风净化笼中饲养。, http://www.100md.com(赵文晓 季旭明 王媛)
