缬草醛对肠易激综合征模型大鼠结肠CRF,TPH1 mRNA及5—HT表达的影响(1)

     [摘要]该研究旨在通过探讨蜘蛛香环烯醚萜类成分缬草醛对肠易激综合征(IBS)大鼠结肠中促肾上腺皮质激素释放因子(CRF)、色氨酸羟化酶1(TPH1) mRNA表達及5羟色胺(5HT)含量的影响,阐释缬草醛通过5HT通路治疗IBS的作用机制。将54只SD 雄性大鼠随机分为空白对照组、模型组、缬草醛高、中、低剂量组、匹维溴铵组,每组9只。采用孤养结合慢急性不可预见性应激法建立IBS大鼠模型,免疫组化法测定大鼠结肠CRF表达,RTPCR法测定结肠中TPH1 mRNA表达,高效液相色谱法测定大鼠结肠中5HT含量。实验结果显示,孤养结合慢急性不可预见性应激可造成大鼠结肠中CRF,TPH1 mRNA表达以及5HT含量升高(P<005),缬草醛各剂量组中CRF,TPH1 mRNA及5HT含量较模型组明显下降(P<005)。实验结果表明,肠易激综合征能够导致结肠中CRF,TPH1 mRNA的表达以及5HT的含量升高,而缬草醛通过降低结肠中CRF,TPH1 mRNA的表达以及5HT的含量,而改善胃肠道的相关症状。

    Effects of baldrinal of Valeriana jatamansi on expression of CRF,TPH1 mRNA and 5HT in rats with irritable bowel syndrome

    [Abstract]This study aimed to investigate the effects of baldrinal of Valeriana jatamansi on the expression of corticotropin releasing factor (CRF) and tryptophan hydroxylase 1 (TPH1) mRNA and levels of 5hydroxytryptamine (5HT) in colon of rats with irritable bowel syndrome (IBS), and to explain its therapeutic mechanism on IBS through 5HT pathway Fiftyfour male SD rats were randomly divided into 6 groups: blank group, model group, baldrinal high, medium and low dose groups, and pinaverium bromide group, n=9 in each group The IBS rat models were established by using unpredictable chronic stress for 3 weeks followed by 1hour acute restraint stress (CAS) after 7 days of rest and independent feeding CRF expression was detected by IHCP; TPH1 mRNA expression was detected by using RTPCR and the 5HT level was measured by high performance liquid chromatography(HPLC) The results indicated that the method of chronic stress with acute restrain stress method and independent feeding could lead to the increase in expressions of CRF and TPH1 mRNA and levels of 5HT in IBS rats(P<005) The expressions of CRF, TPH1 mRNA and 5HT in baldrinal groups were significantly lower than those in model group(P<005) The experimental results showed that IBS could result in increase in the expressions of CRF, TPH1 mRNA and levels of 5HT, and the baldrinal of V jatamansi could improve the symptoms of IBS by reducing the expressions of CRF, TPH1 mRNA and levels of 5HT in colon of rats
