柿叶水提物的抗氧化和抗缺氧作用

     【摘要】 目的 研究柿叶的抗氧化和抗缺氧药理作用。 方法与结果 用NG108-15神经细胞培养过氧化氢(H 2 O 2 )氧化应激和小鼠缺氧模型研究柿叶水提物的抗氧化和抗缺氧药理作用,发现5~100μg/ml柿叶水提物能明显提高H 2 O 2 引起的氧化应激损伤的培养NG108-15神经细胞活力,缓解氧化应激引起的神经细胞内GSH-Px和GSH的减少、MDA的增加以及LDH的漏出。灌胃给予50~300mg/kg柿叶水提物能明显延长小鼠常压缺氧存活时间;延长小鼠亚硝酸钠中毒缺氧存活时间和小鼠急性脑缺血性缺氧的喘气时间。 结论 柿叶水提物有明显的抗氧化和提高缺氧耐受力功能。

    Protection of extract of leaves of diospyros kaki against oxidative stress and hy-poxia

    【Abstract】 Objective To evaluate the protection of water extract of leaves of Diospyros kaki(WELDK)against oxidative stress in vitro and against hypoxia in vivo.Methods NG108-15cell culture was exposed to300μM hydro-gen peroxide(H 2 O 2 )pretreated with/without WELDK.The cell viability was assayed by MTT method.Glutathione(GSH),glutathione peroxidase(GSH-Px),malondialdehyde(MDA)and lactate dehydrogenase(LDH)in the cul-tured cells were assayed using corresponding assay kits.Mice survive time were tested under hypoxia with or without NaNO 2 toxication.Results Pretreating with5~100μg/ml of WELDK was found to raise the cell viability ......
